When you have a twin outboard application is it mandatory to run one left hand rotation, If not how will it affect the boat with two rights?

its not manditory..but having a counter rotation on the portside certainly improves tracking , handling & the feel of your steering.

having twin right handers will make the boat heavier to steer to port & depending on the type of hull ..(& some are affected worse than others) they can create a slightly lent over ride. & a crab walking effect.(tracking).

if you have a choice definatly go for a counter rotating set up.
